The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.

Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
=== Cardiovascular === Like other NSAIDs, its use is associated with an increased risk of cardiovascular events such as heart attack and stroke. Although meloxicam inhibits formation of thromboxane A, it does not appear to do so at levels that would interfere with platelet function. A pooled analysis of randomized, controlled studies of meloxicam therapy of up to 60 days duration found that meloxicam was associated with a statistically significantly lower number of thromboembolic complications than the NSAID diclofenac (0.2% versus 0.8% respectively) but a similar incidence of thromboembolic events to naproxen and piroxicam. People with hypertension, high cholesterol, or diabetes are at risk for cardiovascular side effects. People with family history of heart disease, heart attack, or stroke should tell their treating physician as the potential for serious cardiovascular side effects is significant.
=== Bacteria === Brine pockets are home to a diverse and dynamic community of marine bacteria which are adapted to survive and thrive in the extreme cold, called psychrophiles. As psychrophiles are adapted to survive and grow at very low temperatures, they are capable of synthesizing enzymes that remain active at low temperatures, allowing them to metabolize in the extremely cold conditions of brine pockets and channels. Bacteria in brine pockets must also be able to tolerate high salt concentrations, so these bacteria are also halophilic. Halophilic psychrophiles are found within Proteobacteria, Actinobacteria and Bacteroidetes. Two Proteobacteria found to be abundant in brine pockets are gammaproteobacteria and alphaproteobacteria. Many gammaproteobacteria are capable of degrading organic matter, making them important for nutrient cycling and organic matter turnover within the brine pocket. For example, aerobic anoxygenic phototrophic (AAP) bacteria are found in marine environments and play a vital role in supporting the electron transport chain by metabolizing bacteriochlorophyll. Alphaproteobacteria include species that are known to be important for nitrogen cycling and carbon cycling in marine environments. Some Alphaproteobacteria are capable of nitrogen fixation, which can provide an important source of nitrogen for other microorganisms within the pocket.
The advantages of supercritical fluid extraction (compared with liquid extraction) are that it is relatively rapid because of the low viscosities and high diffusivities associated with supercritical fluids. Alternative solvents to supercritical fluids may be poisonous, flammable or an environmental hazard to a much larger extent than water or carbon dioxide are. The extraction can be selective to some extent by controlling the density of the medium, and the extracted material is easily recovered by simply depressurizing, allowing the supercritical fluid to return to gas phase and evaporate leaving little or no solvent residues. Carbon dioxide is the most common supercritical solvent. It is used on a large scale for the decaffeination of green coffee beans, the extraction of hops for beer production, and the production of essential oils and pharmaceutical products from plants. A few laboratory test methods include the use of supercritical fluid extraction as an extraction method instead of using traditional solvents.
Cardiac troponin C (cTnC) TNNC1 is expressed in cardiac and slow skeletal muscle, while skeletal troponin C (sTnC) TNNC2 is expressed in fast skeletal muscle. sTnC has four calcium ion-binding sites, whereas in cTnC there are only three. The actual amount of calcium that binds to troponin has not been definitively established. Because slow skeletal muscle and myocardium share a type, TnC is not used in diagnostics. Mammals have three troponin I (TnI) genes: the cardiac (TNNI3, cTnI), the slow skeletal (TNNI1), and the fast skeletal (TNNI2). Because the heart uses its own version, blood levels of cTnI is used as a clinical marker. Mammals have three troponin T (TnT) genes: the cardiac (TNNT2, cTnT), the slow skeletal (TNNT1), and the fast skeletal (TNNT3). Again, because the heart uses its own version, cTnT is used as a clinical marker. In addition to the myocardium, there are reports of CTnI and CTnT expression in the wall muscle coat of the vena cava and pulmonary veins, so they do not purely originate from the heart. The expression of cTnT in skeletal tissue of patients with chronic skeletal muscle injuries has been described.
=== Success === SOLAR was known for several others who enjoyed success, including: The Whispers, Dynasty, Lakeside, Midnight Star, Klymaxx, Calloway, Carrie Lucas, Collage and The Deele — which introduced singer/songwriter/producer Kenneth "Babyface" Edmonds and future music executive Antonio "L.A." Reid. Griffey had always believed in giving new talents the opportunity to create and develop their craft, and he was introduced to songwriters/producers Jimmy Jam and Terry Lewis by his A & R rep Dina R. Andrews (Dina Andrews Management Inc) who was the team's first manager, Reggie and Vincent Calloway, and Leon F. Sylvers III. The "SOLAR sound" was a collective effort, with artists working on each other's sessions and artists encouraged to be creative. Sylvers became SOLAR's house producer in 1978 and his signature basslines and productions helped mould the hit sound of SOLAR, which is funky, progressive dance music infused with soul and disco.

The ants plant the seeds and the domatia of young seedling are immediately occupied and the ant faeces in them contribute to rapid growth. Similar dispersal associations are found with other dolichoderines in the region as well. Another example of this type of ectosymbiosis comes from the Macaranga tree, which has stems adapted to house colonies of Crematogaster ants. Many plant species have seeds that are adapted for dispersal by ants. Seed dispersal by ants or myrmecochory is widespread, and new estimates suggest that nearly 9% of all plant species may have such ant associations. Often, seed-dispersing ants perform directed dispersal, depositing the seeds in locations that increase the likelihood of seed survival to reproduction. Some plants in arid, fire-prone systems are particularly dependent on ants for their survival and dispersal as the seeds are transported to safety below the ground. Many ant-dispersed seeds have special external structures, elaiosomes, that are sought after by ants as food. Ants can substantially alter rate of decomposition and nutrient cycling in their nest. By myrmecochory and modification of soil conditions they substantially alter vegetation and nutrient cycling in surrounding ecosystem. A convergence, possibly a form of mimicry, is seen in the eggs of stick insects. They have an edible elaiosome-like structure and are taken into the ant nest where the young hatch.
The normal function of Aβ is not yet known. Though some animal studies have shown that the absence of Aβ does not lead to any obvious loss of physiological function, several potential activities have been discovered for Aβ, including activation of kinase enzymes, protection against oxidative stress, regulation of cholesterol transport, functioning as a transcription factor, and anti-microbial activity (potentially associated with Aβ's pro-inflammatory activity). The glymphatic system clears metabolic waste from the mammalian brain, and in particular amyloid beta. A number of proteases have been implicated by both genetic and biochemical studies as being responsible for the recognition and degradation of amyloid beta; these include insulin degrading enzyme and presequence protease. The rate of removal is significantly increased during sleep. However, the significance of the glymphatic system in Aβ clearance in Alzheimer's disease is unknown.
The perineal body is a pyramidal structure of muscle and connective tissue and part of it is located between the anus and vagina. It is a tendon that is formed at the point where the bulbospongiosus muscle, superficial transverse perineal muscle, and external anal sphincter muscle converge to form this major supportive structure of the pelvis and vagina. Below this, muscles and their fascia converge and become part of the perineal body. The lower vagina is attached to the perineal body by attachments from the pubococcygeus, perineal muscles, and the anal sphincter. The perineal body is made up of smooth muscle, elastic connective tissue fibers, and nerve endings. Above the perineal body are the vagina and the uterus. Damage and resulting weakness of the perineal body changes the length of the vagina and predisposes it to rectocele and enterocele.
MDMA, also known as 3,4-methylenedioxy-N-methylamphetamine or as 1-(1,3-benzodioxol-5-yl)-N-methylpropan-2-amine, is a chemical compound of the phenethylamine, amphetamine, and methylenedioxyphenethylamine (MDxx) families. It is a derivative of various related compounds including phenethylamine, amphetamine, methamphetamine, homopiperonylamine (MDPEA), and 3,4-methylenedioxyamphetamine (MDA). MDMA is a racemic mixture of two enantiomers, (R)-MDMA and (S)-MDMA.
== Structure == Although no crystallographic x-ray diffraction analyses have been published that depict the entire structure of ADAM10, one domain has been studied using this technique. The disintegrin and cysteine-rich domain (shown to the right) plays an essential role in regulation of protease activity in vivo. Recent experimental evidence suggests that this region, which is distinct from the active site, may be responsible for substrate specificity of the enzyme. It is proposed that this domain binds to particular regions of the enzyme's substrate, allowing peptide bond hydrolysis to occur in well defined locations on certain substrate proteins. The proposed active site of ADAM10 has been identified by sequence analysis, and is identical to enzymes in the Snake Venom metalloprotein domain family. The consensus sequence for catalytically active ADAM proteins is HEXGHNLGXXHD. Structural analysis of ADAM17, which has the same active site sequence as ADAM10, suggests that the three histidines in this sequence bind a Zn2+ atom, and that the glutamate is the catalytic residue.

It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
