Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.

Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
==== Enzymes used ==== A commonly used protease mixture is "Flavourzyme", extracted from Aspergillus oryzae, the mold used for soy sauce production. This mixture contains both endo- and exo-peptidases. The endopeptidase Alcalase may also be used, but without an exopeptidase it tends to generate a bitter flavor. As a result, it should be used with a companion exopeptidase. A commercial exopeptidase produced for this purpose is "Protana Prime", a mixture with both leucine aminopeptidase and carboxypeptidase D activity. Beyond proteolysis, the amount of umami taste can also be increased by adding a glutaminase, which converts glutamine to glutamate. Commercial options include "Protana Boost" and others.
== Environmental impact and regulation == PFBA is a breakdown product of other PFAS that have been used in stain-resistant fabrics, paper food packaging, carpets, and consumer products. PFBA has been frequently found in U.S. rivers. In laboratory animal studies, exposure to high levels of PFBA results in thyroid and liver effects, such as increased thyroid and liver weight, changes in thyroid hormones, decreased cholesterol, and cellular changes in both organs. Other effects of PFBA exposure included delayed development and decreased red blood cells and hemoglobin. The Minnesota Department of Health (MDH) developed a guidance value of 7 ppb (μg/L) for PFBA in drinking water.
==== 2011 Veracruz massacres ==== In spring 2011, the CJNG declared war on all other Mexican cartels and stated its intention to take control of the city of Guadalajara. However, by midsummer, the group appeared to have been reunited with its former partners in the Sinaloa Cartel. In addition to maintaining its anti-Zetas alliance with the Gulf Cartel, the Sinaloa Cartel in 2011 affiliated itself with the Knights Templar in Michoacán. To counter Los Zetas in the state of Jalisco, the Sinaloa Cartel affiliated itself with the CJNG. On 20 September 2011, two trucks containing 35 dead bodies were found at an underpass near a shopping mall in Boca del Río, Veracruz. All of the corpses were alleged to be members of Los Zetas, but it was later proven that only six of them had been involved in minor crime incidents, and none of them were involved with organized crime. Some of the victims had their hands tied and showed signs of having been tortured. According to El Universal, at around 17:00 an undetermined number of vehicles blocked a major avenue in Boca del Río. Once the traffic stopped, armed men abandoned two trucks in the middle of the highway. They opened the doors of the trucks and pulled out the thirty-five corpses, leaving a written message behind. Other gunmen pointed their weapons at the frightened drivers. The gunmen then fled the scene. Consequently, the stunned motorists began to grab their cellphones and post messages on Twitter warning other drivers to avoid the area. The message left behind stated the following:
==== Photografting ==== The modification of inert surfaces of polyolefins, polyesters, and polyamides by grafting functional vinyl monomers has been used to increase hydrophobicity, dye absorption, and polymer adhesion. This photografting method is generally used during continuous filament or thin film processing. On a bulk commercial scale, the grafting technique is referred to as photoinitiated lamination, where desired surfaces are joined by grafting a polymeric adhesion network between the two films. The low adhesion and absorption of polyolefins, polyesters, and polyamides is improved by UV-irradiation of an initiator and monomer transferred through the vapor phase to the substrate. Functionalization of porous surfaces have seen great success with high temperature photografting techniques. In microfluidic chips, functionalizing channels allows directed flow to preserve lamellar behavior between and within junctions. The adverse turbulent flow in microfluidic applications can compound component failure modes due to the increased level of channel interdependency and network complexity. In addition, the imprinted design of microfluidic channels can be reproduced for photografting the corresponding channels with a high degree of accuracy.

18-Methoxycoronaridine (18-MC; developmental code name MM-110), also known as zolunicant (INNTooltip International Nonproprietary Name), is a derivative of ibogaine invented in 1996 by the research team around the pharmacologist Stanley D. Glick from the Albany Medical College and the chemists Upul K. Bandarage and Martin E. Kuehne from the University of Vermont. 18-MC was originally developed by Savant HWP and later acquired by MindMed in 2019 for development as a treatment for opioid use disorder. A Phase 1 trial in healthy volunteers was completed in 2022 with favorable safety and tolerability. Due to strategic reprioritization, MindMed discontinued active development of MM-110 in 2023 and has been seeking non-dilutive funding or partners to potentially restart the program; as of 2025 the program remains shelved. A separate Phase 2 trial in Brazil for cutaneous leishmaniasis (initiated 2017) has unknown status with no published results.

KFC's core product offering is pressure fried on-the-bone chicken pieces seasoned with Colonel Harland Sanders' "Original Recipe" of 11 herbs and spices. The product is typically available in either two- or three-piece individual servings or in a family size cardboard bucket typically holding between six and 16 chicken pieces. In territories that follow the system handed down by Colonel Sanders, such as Canada and the UK, each chicken is divided into nine different cuts (two drumsticks, two thighs, two wings, two breast pieces and one keel); however, the United States now uses an eight-piece cut. The chicken is hand-breaded at individual KFC outlets with wheat flour mixed with seasoning in a two- to four-minute process. It is then pressure fried for between seven and 10 minutes (the timing differs between countries) in oil at 185 degrees Celsius. Following this, the chicken is left to stand for 5 minutes in order for it to sufficiently cool before it is placed in the warming oven. It is KFC policy to discard chicken if it has not been sold within 90 minutes in order to ensure freshness. The frying oil varies regionally and versions used include sunflower, soybean, rapeseed and palm oil. A KFC executive stated that the taste of the chicken will vary between regions depending on the oil variety used and whether the chicken has been corn-fed or wheat-fed.
The earliest study of the nervous system dates to ancient Egypt. Trepanation, the surgical practice of either drilling or scraping a hole into the skull for the purpose of curing head injuries or mental disorders, or relieving cranial pressure, was first recorded during the Neolithic period. Manuscripts dating to 1700 BC indicate that the Egyptians had some knowledge about symptoms of brain damage. Early views on the function of the brain regarded it to be a "cranial stuffing" of sorts. In Egypt, from the late Middle Kingdom onwards, the brain was regularly removed in preparation for mummification. It was believed at the time that the heart was the seat of intelligence. According to Herodotus, the first step of mummification was to "take a crooked piece of iron, and with it draw out the brain through the nostrils, thus getting rid of a portion, while the skull is cleared of the rest by rinsing with drugs." Greek thinkers, beginning with Alcmaeon of Croton, advanced the cephalocentric hypothesis that the brain is the seat of sensation and thought. The view that the heart was the source of consciousness was not challenged until the time of the Greek physician Hippocrates. He believed that the brain was not only involved with sensation—since most specialized organs (e.g., eyes, ears, tongue) are located in the head near the brain—but was also the seat of intelligence. Plato also speculated that the brain was the seat of the rational part of the soul.
Portal received critical acclaim, often earning more praise than either Half-Life 2: Episode Two or Team Fortress 2, two titles also included in The Orange Box. It was praised for its unique gameplay and dark, deadpan humor. Eurogamer cited that "the way the game progresses from being a simple set of perfunctory tasks to a full-on part of the Half-Life story is absolute genius", while GameSpy noted, "What Portal lacks in length, it more than makes up for in exhilaration." The game was criticized for sparse environments, and both criticized and praised for its short length. Aggregate reviews for the standalone PC version of Portal gave the game a 90/100 through 28 reviews on Metacritic. In 2011, Valve stated that Portal had sold more than four million copies through the retail versions, including the standalone game and The Orange Box, and from the Xbox Live Arcade version. The game generated a fan following for the Weighted Companion Cube—even though the cube itself does not talk or act in the game. Fans have created plush and papercraft versions of the cube and the various turrets, as well as PC case mods and models of the Portal cake and portal gun. Jeep Barnett, a programmer for Portal, noted that players have told Valve that they had found it more emotional to incinerate the Weighted Companion Cube than to harm one of the "Little Sisters" from BioShock.

Long-term storage is usually at minus 20 to minus 80 degrees Celsius in a sealed, desiccated container. Working aliquots can be held briefly at 2 to 8 degrees Celsius.
Repeated temperature cycling can promote aggregation and peptide degradation. Dividing material into single-use aliquots limits this risk.
Mass spectrometry is commonly used to confirm molecular mass and detect structural modifications. It is often paired with chromatographic purity assessment.
It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
