Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.

Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
[A]bout one-third of the Gratiot-area's families eventually moved to public housing, but 35 percent of the families in the area could not be traced. The best-informed city officials believed that a majority of families moved to neighborhoods within a mile of the Gratiot site, crowding into an already decaying part of the city, and finding houses scarcely better and often more overcrowded than that which they had left.
Aerobic exercise comprises innumerable forms. In general, it is performed at a moderate level of intensity over a relatively long period of time. For example, running a long distance at a moderate pace is an aerobic exercise, but sprinting is not. Playing singles tennis, with near-continuous motion, is generally considered aerobic activity, while activities with brief bursts of energetic movement within longer periods of casual movement may not be aerobic. Some sports are thus inherently "aerobic", while other aerobic exercises, such as fartlek training or aerobic dance classes, are designed specifically to improve aerobic capacity and fitness. It is most common for aerobic exercises to involve the leg muscles, primarily or exclusively. There are some exceptions. For example, rowing to distances of 2,000 meters or more is an aerobic sport that exercises several major muscle groups, including those of the legs, abdominals, chest, and arms.
Annealing: This is a thermal treatment generally used to soften the material being drawn; to modify the microstructure, the mechanical properties, and the machining characteristics of the steel; and to remove internal stresses in the product. Depending on the material and desired final characteristics, annealing may be used before, during (between passes), or after the cold drawing operation.

== BDNF-AS == The genomic locus encoding BDNF is structurally complex and also encodes BDNF-antisense (BDNF-AS; also known as BDNFOS or ANTI-BDNF). BDNF-AS is a long non-coding RNA (lncRNA) transcribed from the opposite strand of the BDNF gene. This lncRNA was identified in 2005 through searches in expressed sequence tag (EST) databases and subsequent RT-PCR experiments. The gene encoding BDNF-AS is located on chromosome 11p14.1. BDNF mRNA and BDNF-AS share a common overlapping region and form double-stranded RNA (dsRNA) duplexes. BDNF-AS regulates BDNF expression and can suppress BDNF mRNA. In the human neocortex, regions with increased activity and BDNF expression exhibit reduced BDNF-AS expression. Elevated BDNF-AS levels are associated with reduced BDNF expression and have been shown to promote neurotoxicity, increase apoptosis, and decrease cell viability. Conversely, inhibiting BDNF-AS upregulates BDNF mRNA, activates BDNF-mediated signaling pathways, increases BDNF protein levels, suppresses neuronal apoptosis, and promotes neuronal outgrowth and differentiation. The BDNF-AS gene consists of 10 exons and a functional promoter upstream of exon 1. The BDNF-AS gene generates numerous distinct non-coding RNAs through alternative splicing. This diversity of spliced isoforms is a common feature of eukaryotic organisms, particularly in the nervous system. Notably, BDNF-AS is absent in rodents, although highly homologous sequences are present in the genomes of chimpanzees and rhesus monkeys, suggesting a primate/hominid evolutionary origin of BDNF-AS.
Tacrolimus and a related drug for eczema (pimecrolimus) were suspected of carrying a cancer risk, though the matter is still a subject of controversy. The FDA issued a health warning in March 2005 for the drug, based on animal models and a small number of patients. Until further human studies yield more conclusive results, the FDA recommends that users be advised of the potential risks. However, current practice by UK dermatologists is not to consider this a significant real concern and they are increasingly recommending the use of these new drugs. A 2023 systematic review and meta-analysis published in The Lancet Child & Adolescent Health concluded with moderate-certainty evidence that the two drugs were not associated with any increased risk of cancer. In November 2024, International Agency for Research on Cancer (IARC) classified hydrochlorothiazide, voriconazole and tacrolimus as group 1 carcinogens.
For some individuals who experience difficulty in achieving full glanular engorgement of glans penis, they may be diagnosed with soft glans syndrome (glans insufficiency syndrome). It is often undiagnosed in the general population due to the lack of a standardized nomenclature.
== Veterinary use == It can be used for treating infected wounds on animals. Usually in powder form mixed with water, it has a color and smell similar to Tang. Given orally to animals, the amount is dependent on their weight and severity of infection.
==== South Africa ==== There are two major brands of cider produced in South Africa, Hunters and Savanna. They are produced and distributed through Heineken, which acquired Distell in April 2023. Hunters Gold was first introduced in South Africa in 1988 as an alternative to beer. The Hunters range includes Hunters Dry, Hunters Gold, Hunters Export and Hunters Edge launched in April 2017. Savanna Dry was introduced in 1996 and also comes in Light, Angry Lemon and Alc Free variants, excluding other limited run variants like Loco, Blackbeard and Neat. The Elgin Valley east of Cape Town grows around 60% of all the apples in South Africa. As a result, the region also has a vibrant craft cider scene, with brands like Sxollie, Everson's, Cluver & Jack, Windermere and Terra Madre all produced in the area. Sxollie Cider is increasingly being exported around the world, especially to the UK. Almost 90% of all the Sxollie produced is exported. Sxollie is pronounced like "skollie". Sxollie's Pink Lady Cider won Supreme Champion at the International Cider Challenge in 2019. Windermere Cider began brewing in 1994 and is regarded as the oldest craft cider brewery in South Africa. Loxtonia Cider, produced in the Ceres area, was crowned the overall champion at the 11th International Cider Challenge in London in 2021. Loxtonia's Alexandra Blush Méthode Traditionelle was named Supreme Champion for 2021 and won the Rosé Trophy as the best cider made in the rosé style.

== Binding specificity == The effects of a drug are a result of their binding selectivity with macromolecule properties of an organism, or the affinity with which different ligands bind to a substrate. More specifically, the specificity and selectivity of a ligand to its respective receptor provides researchers the opportunity to isolate and produce specific drug effects through the manipulation of ligand concentrations and receptor densities. Hormones and neurotransmitters are essential endogenous regulatory ligands that affect physiological receptors within an organism. Drugs that act upon these receptors are incredibly selective in order to produce required responses from signaling molecules. Specific binding refers to the binding of a ligand to a receptor, and it is possible that there is more than one specific binding site for one ligand. Non specific binding refers to the binding of a ligand to something other than its designated receptor such as various other receptors, or different types of transporters in the cell membrane. For example, various antagonists can bind to multiple types receptors. In the case of muscarinic antagonists, they can also bind to histamine receptors. Such binding patterns are technically considered specific, as the destination of the ligand is specific to multiple receptors. However, researchers may not be focused on such behaviors compared to other binding factors. Nevertheless, nonspecific binding behavior is very important information to acquire.
Sniper is a series of action and war films beginning with the 1993 film Sniper, which center upon the characters of Master Gunnery Sergeant Thomas Beckett (Tom Berenger) and Gunnery Sergeant Brandon Beckett (Chad Michael Collins), who work as Force Reconnaissance Scout Snipers in the United States Marine Corps.
Pressure ulcers, also known as pressure sores, bed sores or pressure injuries, are localised damage to the skin and/or underlying tissue that usually occurs over a bony prominence as a result of usually long-term pressure, or pressure in combination with shear or friction. The most common sites are the skin overlying the sacrum, coccyx, heels, and hips, though other sites can be affected, such as the elbows, knees, ankles, back of shoulders, or the back of the cranium. Pressure ulcers occur due to pressure applied to soft tissue which results in completely or partially obstructed blood flow to the soft tissue. Shear is also a cause, as it can pull on blood vessels that feed the skin. Pressure ulcers most commonly develop in individuals who are not moving about, such as those who are on chronic bedrest or consistently use a wheelchair. It is widely believed that other factors can influence the tolerance of skin for pressure and shear, thereby increasing the risk of pressure ulcer development. These factors are protein-calorie malnutrition, microclimate (skin wetness caused by sweating or incontinence), diseases that reduce blood flow to the skin, such as arteriosclerosis, or diseases that reduce the sensation in the skin, such as paralysis or neuropathy. The healing of pressure ulcers may be slowed by a person's age, medical conditions (such as arteriosclerosis, diabetes or infection), smoking or medications such as anti-inflammatory drugs.
These reactions are divided into "cold" and "hot" fusion, which respectively create systems with lower and higher excitation energies; this affects the yield of the reaction. For example, the reaction between 248Cm and 40Ar was expected to yield isotopes of element 114, and that between 232Th and 84Kr was expected to yield isotopes of element 126. None of these attempts were successful, indicating that such experiments may have been insufficiently sensitive if reaction cross sections were low—resulting in lower yields—or that any nuclei reachable via such fusion-evaporation reactions might be too short-lived for detection. Subsequent successful experiments reveal that half-lives and cross sections indeed decrease with increasing atomic number, resulting in the synthesis of only a few short-lived atoms of the heaviest elements in each experiment; as of 2022, the highest reported cross section for a superheavy nuclide near the island of stability is for 288Mc in the reaction between 243Am and 48Ca. Similar searches in nature were also unsuccessful, suggesting that if superheavy elements do exist in nature, their abundance is less than 10−14 moles of superheavy elements per mole of ore. Despite these unsuccessful attempts to observe long-lived superheavy nuclei, new superheavy elements were synthesized every few years in laboratories through light-ion bombardment and cold fusion reactions; rutherfordium, the first transactinide, was discovered in 1969, and copernicium, eight protons closer to the island of stability predicted at Z = 114, was reached by 1996.

It is a modified version of the natural hormone, with three amino acid changes and a fatty acid side chain added. These edits extend its half-life from minutes to about one week. The core receptor activity is retained.
