Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.

The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.
Small humanin-like peptides (SHLPs) are a group of peptides encoded in the 16S ribosomal RNA region of mitochondrial genome. Six peptides of this group (SHLP1–6) have been identified so far, between 20-38 amino acids in length. They are derived from the mitochondria and act as important retrograde signaling molecules in the cell. Their names were given because the SHLPs share some similar biological effects with the mitochondrial peptide Humanin. Every individual SHLP showed unique expression pattern in mouse tissues. Specifically, SHLP1 was detected in the heart, kidney, and spleen; SHLP2 was detected in the liver, kidney, and muscle; SHLP3 was detected in the brain and spleen; SHLP4 was detected in the liver and prostate; and SHLP6 was detected in the liver and kidney. Experiments using cultured mammalian cells have shown that SHLPs are bioactive peptides. Incubation of each synthetic SHLP with cells affected cell viability, proliferation and apoptosis differentially, which suggests that every SHLP may play a different role in the biological system.
The thoracic cavity is protected by the thoracic wall (comprising the rib cage and associated skin, muscle, and fascia), comprising the mediastinum, heart and great vessels, and the bulk of the respiratory tract. The abdominal cavity, lined by the peritoneum, contains the bulk of the gastrointestinal tract, the spleen and the kidneys. Abdominal organs may be classified as solid organs or hollow organs. The solid organs are the liver, pancreas, spleen, kidneys, and adrenal glands. The hollow organs of the abdomen are the stomach, intestines, gallbladder, bladder, and rectum. The pelvic cavity contains the urinary bladder, internal reproductive organs, and rectum. Viscera are the internal organs of the ventral cavity. The term "visceral" is contrasted with the term "parietal", meaning "of or relating to the wall of a body part, organ or cavity". The two terms are often used in describing a membrane or piece of connective tissue, referring to the opposing sides.
According to the United Nations Office on Drugs and Crime World Drug Report, approximately 21 million people worldwide use ecstasy-type substances, including MDMA. This accounts for roughly 0.4% of the global population aged 15 to 64. In the United States, according to the Substance Abuse and Mental Health Services Administration 2021 National Survey on Drug Use and Health, among people aged 12 or older, 0.8% (2.2 million people) reported the use of ecstasy in the past year. Usage is much higher among attendees of nightclubs and dance festivals and among people aged in their late 20s.
Histidine is the precursor to histamine. Tryptophan is the precursor to 5-hydroxytryptophan and then serotonin, tryptamine, auxin, kynurenines, and melatonin. Tyrosine is the precursor to L-DOPA, dopamine, norepinephrine (noradrenaline), epinephrine (adrenaline), and the thyroid hormone thyroxine. It is also precursor to octopamine and melanin in numerous organisms. Phenylalanine is the precursor to tyrosine.
In 1978 he had established a Women's Military Academy in Tripoli, encouraging all women to enlist for training. The measure was hugely controversial and voted down by the GPC in February 1983. Gaddafi remained adamant, and when it was again voted down by the GPC in March 1984, he refused to abide by the decision, declaring that "he who opposes the training and emancipation of women is an agent of imperialism, whether he likes it or not." The Jamahiriya's radical direction earned the government many enemies. Most internal opposition came from Islamic fundamentalists, inspired by the events of the 1979 Iranian Revolution. In February 1978, Gaddafi discovered that his head of military intelligence was plotting to kill him and increasingly entrusted security to his Qadhadfa tribe. Many who had seen their wealth confiscated turned against the administration, and Western-funded opposition groups were founded by exiles. Most prominent was the National Front for the Salvation of Libya (NFSL), which orchestrated militant attacks against Libya's government. Another, al-Borkan, began killing Libyan diplomats abroad. Following Gaddafi's command to kill these "stray dogs", the Revolutionary Committees set up overseas branches to suppress counter-revolutionary activity, assassinating dissidents. Although Syria and Israel also employed hit squads, Gaddafi was unusual in publicly bragging about his use of them; in April 1980, he ordered all dissidents to return home by 10 June or be "liquidated wherever you are".

=== Pioneering hydroxamate structures === The first generation of MMP inhibitors were based on the structure of the collagen molecule. In the design of these inhibitors, the basic protein backbone of collagen is maintained but the amide bond is replaced with a zinc-binding group. This group of inhibitors contain a hydroxamate (-CONHOH) group that binds the zinc atom in the active site of the MMP enzyme, therefore this group is called "hydroxamate-based MMP inhibitors“. An example can be seen in Marimastat, a first generation inhibitor, which has a similar backbone and sidechain format to collagen.
because it is above thorium's 6.3 eV first ionization energy, neutral 229mTh can decay by internal conversion, and will almost always do so, rather than emitting a photon, and because it is above the 6.2 eV vacuum ultraviolet cutoff, the produced photons cannot travel through air. But even when experiments were redesigned to detect the higher energy, most of the searches in the 2010s for light emitted by the isomeric decay failed to observe any signal, pointing towards a potentially strong non-radiative decay channel. A direct detection of photons emitted in the isomeric decay was claimed in 2012 and again in 2018. However, both reports were controversial within the community. A direct detection of electrons being emitted in the internal conversion decay channel of 229mTh was achieved in 2016. However, at the time the isomer's transition energy could only be weakly constrained to between 6.3 and 18.3 eV. Finally, in 2019, non-optical electron spectroscopy of the internal conversion electrons emitted in the isomeric decay allowed for a determination of the isomer's excitation energy to 8.28±0.17 eV. However, this value appeared at odds with the 2018 preprint showing that a similar signal as an 8.4 eV xenon VUV photon can be shown, but with about 1.3+0.2−0.1 eV less energy and a (retrospectively correct) 1880±170 s lifetime. In that paper, 229Th was embedded in SiO2, possibly resulting in an energy shift and altered lifetime, although the states involved are primarily nuclear, shielding them from electronic interactions.
== Pharmacology == Like ibogaine, it has seems to have similar pharmacology. It has effects on KOR, NMDAR, nAChR and serotonin sites. It also inhibits acetylcholinesterase and butyrylcholinesterase. Ibogamine persistently reduced the self-administration of cocaine and morphine in rats. The same study found that ibogamine (40 mg/kg) and coronaridine (40 mg/kg) did not produce "any tremor effects in rats that differ significantly from saline control". While the related alkaloids ibogaine (20–40 mg/kg), harmaline (10–40 mg/kg) and desethylcoronaridine (10–40 mg/kg) were "obviously tremorgenic".

=== Elder welfare === In 2004, Ambani established Harmony for Silvers Foundation, a Mumbai-based non-government organisation that seeks to enhance the quality of life of the elderly. Its activities have included Harmony – Celebrate Age, the magazine, now in its 14th year; the portal www.harmonyindia.org; Harmony Interactive Centre for Silver Citizens, in South Mumbai; the Harmony Research Division; the Harmony Silver Awards; and the Harmony Senior Citizens' Runs at the Mumbai, Delhi and Bengaluru marathons.
Chlorambucil, sold under the brand name Leukeran among others, is a chemotherapy medication used to treat chronic lymphocytic leukemia (CLL), Hodgkin lymphoma, and non-Hodgkin lymphoma. It is given by mouth. Common side effects include bone marrow suppression. Other serious side effects include an increased long term risk of further cancer, infertility, and allergic reactions. Use during pregnancy often results in harm to the baby. Chlorambucil is in the alkylating agent family of medications. It works by blocking the formation of DNA and RNA. Chlorambucil was approved for medical use in the United States in 1957. It is on the World Health Organization's List of Essential Medicines. It was originally made from nitrogen mustard.
Where the concentration of the enzyme is much less than the concentration of the substrate, the rate of reaction is directly proportional to the total enzyme concentration. The hydrolysis of a peptide bond however is an energetically favourable reaction. Recent experiments with both crude caricain and purified caricain indicated that the reaction which controls detoxification of a wheat gliadin digest at pH7.5 and 37 °C was indeed a 1st order reaction with a rate constant of 1.7 x 10 −4 sec.−1. The rate of reaction was followed by the disappearance of gliadin peptides which were toxic to rat liver lysosomes.
=== Internal standards === Sometimes an internal standard is added at a known concentration directly to an analytical sample to aid in quantitation. The amount of analyte present is then determined relative to the internal standard as a calibrant. An ideal internal standard is an isotopically enriched analyte which gives rise to the method of isotope dilution.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
