In 2004, clinical development of CAT-354 was initiated with this first study completing in 2005. On 21 July 2011, MedImmune LLC initiated a Phase IIb, randomized, double-blind study to evaluate the efficacy of tralokinumab in adults with asthma. In 2016, MedImmune and AstraZeneca started developing tralokinumab for asthma (Phase III) and atopic dermatitis (Phase IIb) while clinical development for moderate-to-severe ulcerative colitis and idiopathic pulmonary fibrosis (IPF) have been discontinued. In July of that year AstraZeneca licensed tralokinumab to Leo Pharma for skin diseases. A phase IIb study of tralokinumab found that treatment was associated with early and sustained improvements in atopic dermatitis symptoms and tralokinumab had an acceptable safety and tolerability profile, thereby providing evidence for targeting IL-13 in patients with atopic dermatitis. === Laminitis === EMS is also implicated in the development of laminitis. Prolonged IV insulin administration can induce laminitis, possibly due to its effects on blood flow to the foot, changes of glucose metabolism and secondary matrix metalloproteinase activation, or altered cell function within the foot. However, recent research suggests that the situation is more complex, in that "compensated insulin resistance is essentially physiological and health sustaining", and only when this compensatory mechanism fails does laminitis ensue. This may support the argument that EMS is an evolved survival trait.

Amanita phalloides ( AM-ə-NY-tə fə-LOY-deez), commonly known as the death cap, is a deadly poisonous basidiomycete fungus and mushroom, one of many in the genus Amanita. Originating in Europe but later introduced to other parts of the world since the late twentieth century, A. phalloides forms ectomycorrhizae with various broadleaved trees. In some cases, the death cap has been introduced to new regions with the cultivation of non-native species of oak, chestnut, and pine. The large fruiting bodies appear in summer and autumn; the caps are generally greenish in colour with a white stipe and gills. The cap colour is variable, including white forms, and is thus not a reliable identifier. The death cap resembles several edible species (most notably Caesar's mushroom and the straw mushroom) commonly consumed by humans, increasing the risk of accidental poisoning. Amatoxins, a class of toxins found in mushrooms like A. phalloides, are thermostable: they resist changes due to heat and cold, so their toxic effects are not reduced by cooking or freezing. Amanita phalloides is the most poisonous of all known mushrooms. It is estimated that as little as half a mushroom contains enough toxin to kill an adult human. It is also the deadliest mushroom worldwide, responsible for 90% of mushroom-related fatalities every year. It has been involved in the majority of human deaths from mushroom poisoning, possibly including Roman Emperor Claudius in AD 54 and Holy Roman Emperor Charles VI in 1740.
=== RNAi therapeutics === Just after Fire and Mello's ground-breaking discovery, Elbashir et al. discovered, by using synthetically made siRNA, it was possible to target the silencing of specific sequences in a gene, rather than silencing the entire gene. Only a year later, McCaffrey and colleagues demonstrated that this sequence-specific silencing had therapeutic applications by targeting a sequence from the Hepatitis C virus in transgenic mice. Since then, multiple researchers have been attempting to expand the therapeutic applications of RNAi, specifically looking to target genes that cause various types of cancer. By 2006, the first applications to reach clinical trials were in the treatment of macular degeneration and respiratory syncytial virus. Four years later the first-in-human Phase I clinical trial was started, using a nanoparticle delivery system to target solid tumors. The FDA approved the first siRNA-based drug (patisiran) in 2018. Givosiran and lumasiran later won FDA approval for the treatment of AHP and PH1 in 2019 and 2020, respectively. Inclisiran received EMA approval in 2020 for the treatment of high cholesterol and is currently under review by the FDA.

By the end of August 1944 the German forces in Italy had formed the Gothic Line along the Apennine Mountains, and the 6th Division had come under the command of the US Fifth Army. The difficulty of using tanks in the mountains led to the Rhodesians of Prince Alfred's Guard temporarily adopting an infantry role, using dismounted tank machine-guns to support the Natal Carabineers during the fighting for Pistoia during early September. The Southern Rhodesian Anti-Tank Battery meanwhile converted partially from guns to 4.2-inch mortars. The South Africans and Rhodesians met with fierce resistance from the 16th SS Panzer Grenadier Division, but helped push the Germans northwards towards the Reno river. Hoping to repel Allied advances towards Bologna, the Germans took up positions on Monte Stanco overlooking the main roads towards the city. Two Allied assaults on the mountain—one by an Indian battalion, the other by the Royal Natal Carabineers—were repulsed. A third, larger attack at dawn on 13 October provided the Rhodesian Company of the Cape Town Highlanders with some of the fiercest combat they encountered in Italy. Advancing up the slope on the Allied right flank while being fired on from two directions, they suffered heavy casualties but achieved their objective and held it. Both Rhodesian artillery batteries provided support during the assault. When the line stabilised in November 1944, the portion occupied by the 6th Armoured Division extended for 16 kilometres (9.9 mi) along the heights over the Reno River.

Diacetyldihydromorphine (also known as Paralaudin, dihydroheroin, acetylmorphinol) is a potent opiate derivative developed in Germany in 1928 which is rarely used in some countries for the treatment of severe pain such as that caused by terminal cancer, as another form of diacetylmorphine (also commonly known as Heroin). Diacetyldihydromorphine is fast-acting and longer-lasting than diamorphine, with a duration of action of around 4–7 hours. As an ester/analogue of dihydromorphine, diacetyldihydromorphine is presumably a Schedule I/Narcotic controlled substance in the United States but does not have its own ACSCN or annual production quota. It does appear in the German Betäubungsmittelgesetz and other European controlled-substances laws. Diacetyldihydromorphine is quickly metabolized by plasma esterase enzymes into dihydromorphine, in the same way that diamorphine is metabolized into morphine. Diacetyldihydromorphine is roughly equipotent to morphine, where as diamorphine (heroin) is 1.50–1.80 times the potency of morphine. It shares with other opioids the risk of overdose or (potentially life-threatening) respiratory depression. When strong narcotics are required, and morphine and diamorphine are not an option, it is more common to use better known drugs such as nicomorphine, hydromorphone, levorphanol, oxymorphone or fentanyl which doctors will be more familiar with, and which do not share the stigma associated with either heroin or morphine.
