GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Storage conditions for semaglutide depend heavily on the presentation. Lyophilized research powder is generally kept at two to eight degrees Celsius in a sealed container, protected from light and moisture. Manufacturer labeling for finished injectable products specifies refrigeration before first use, with defined in-use periods at room temperature afterward. The oral tablet form is stored at controlled room temperature and is more tolerant of short excursions. Temperature excursions should be documented rather than inferred.
Peptide degradation follows several routes. Hydrolysis cleaves the backbone at susceptible residues, oxidation targets methionine and tryptophan side chains, and aggregation produces higher-molecular-weight species that are difficult to reverse. Light exposure accelerates oxidation, which is why amber glass or opaque secondary packaging is common. Repeated freeze-thaw cycles promote aggregation and are best avoided. Stability-indicating methods detect these changes before they become visible.
Quality control for research material typically involves reversed-phase HPLC for purity and identity, mass spectrometry for molecular weight confirmation, and Karl Fischer titration for residual water content. Peptide content is often reported as the mass of actual peptide rather than total powder mass, since counterions and water contribute to the latter. A certificate of analysis should list the method used for each specification. Limits and acceptance criteria vary by supplier and by intended application.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Psilocybin is used as a psychedelic at doses of 5 to 40 mg orally. Low doses are 5 to 10 mg, an intermediate or "good effect" dose is 20 mg, and high or ego-dissolution doses are 30 to 40 mg. Psilocybin's effects can be subjectively perceived at a dose as low as 3 mg per 70 kg body weight. Microdosing involves the use of subthreshold psilocybin doses of less than 2.5 mg. When psilocybin is used in the form of psilocybin-containing mushrooms, microdoses are 0.1 g to 0.3 g and psychedelic doses are 1.0 g to 3.5–5.0 g in the case of dried mushrooms. The preceding 1.0 to 5.0 g range corresponds to psilocybin doses of about 10 to 50 mg. Psilocybin-containing mushrooms vary in their psilocybin and psilocin content, but are typically around 1% of the dried weight of the mushrooms (in terms of total or combined psilocybin and psilocin content). Psilocin is about 1.4 times as potent as psilocybin because of the two compounds' difference in molecular weight. "Lemon tek" or "lemon tekking" is a method sometimes used by recreational psilocybin users. It involves soaking psilocybin-containing mushrooms in citric acid-containing lemon juice to supposedly convert their psilocybin content into psilocin before administration. This is claimed to hasten their onset, cause a sharper and more intense peak, and shorten their duration.
Following Maduro's capture, demonstrations celebrating his removal and opposing his presidency occurred in Caracas, the capital and largest city of Venezuela. Venezuelans living abroad celebrated Maduro's removal across the United States, Spain, and Latin America. In Doral, Florida, where the largest population of Venezuelans in the US live, Venezuelans filled the streets waving flags, singing, and chanting "freedom". Thousands of Venezuelans celebrated Maduro's fall in Madrid, Spain. Videos from the Colombian border city of Cúcuta showed crowds filling the streets on the night of 3 January 2026, dancing to music and chanting slogans like "virtue" and "a free Venezuela". Similar celebrations by Venezuelan expatriates and refugees took place in Buenos Aires in Argentina, Lima in Peru, Quito in Ecuador, and Santiago in Chile.
Specially, carbon nanotubes can be transformed into sophisticated biomolecule and allow its detection through changes in the carbon nanotube fluorescence spectra. Also, carbon nanotubes can be designed to match the size of small drug and endocitozed by a target cell, hence becoming a delivery agent.
== Overview == Dating analyses in 2016 determined that Denny (Denisova 11) died about 90,000 years ago, and the bone's cortical thickness indicates an age at death of at least 13 years. An analysis of the whole genome sequence (total mitochondrial and nuclear DNA) indicates she was female, with a Neanderthal mother and a Denisovan father. While previous analyses of other ancient genomes concluded that Denisovans, Neanderthals, and modern humans interbred during the ice age in Europe and Asia, this find is the most direct evidence yet that various ancient hominins mated with each other and had offspring. Previous analyses from other fossils found in this Siberian cave have shown that modern humans, Neanderthals, and Denisovans inhabited this site at various times, and that all three human species interbred with each other. The genes of both archaic human species are present in many people today, which suggests that when these groups met, gene flow occurred. It is not evident if the mating was consensual or if Denny was fertile. The discovery of Denisova 11 may support the notion that Neanderthals and Denisovans may not have undergone direct extinction but were partly assimilated into modern human populations.

Cirrhosis is most commonly caused by medical conditions including alcohol-related liver disease, metabolic dysfunction–associated steatohepatitis (MASH – the progressive form of metabolic dysfunction–associated steatotic liver disease, previously called non-alcoholic fatty liver disease or NAFLD), heroin abuse, chronic hepatitis B, and chronic hepatitis C. Chronic heavy drinking can cause alcoholic liver disease. Liver damage has also been attributed to heroin usage over an extended period of time as well. MASH has several causes, including obesity, high blood pressure, abnormal levels of cholesterol, type 2 diabetes, and metabolic syndrome. Less common causes of cirrhosis include autoimmune hepatitis, primary biliary cholangitis, and primary sclerosing cholangitis that disrupts bile duct function, genetic disorders such as Wilson's disease and hereditary hemochromatosis, and chronic heart failure with liver congestion. Diagnosis is based on blood tests, medical imaging, and liver biopsy. Hepatitis B vaccine can prevent hepatitis B and the development of cirrhosis from it, but no vaccination against hepatitis C is available. No specific treatment for cirrhosis is known, but many of the underlying causes may be treated by medications that may slow or prevent worsening of the condition. Hepatitis B and C may be treatable with antiviral medications. Avoiding alcohol is recommended in all cases. Autoimmune hepatitis may be treated with steroid medications. Ursodiol may be useful if the disease is due to blockage of the bile duct.
== Biological role == Thaumatin production is induced in katemfe in response to an attack upon the plant by viroid pathogens. Several members of the thaumatin protein family display significant in vitro inhibition of hyphal growth and sporulation by various fungi. The thaumatin protein is considered a prototype for a pathogen-response protein domain. This thaumatin domain has been found in species as diverse as rice and Caenorhabditis elegans. Thaumatins are pathogenesis-related (PR) proteins, which are induced by various agents ranging from ethylene to pathogens themselves, and are structurally diverse and ubiquitous in plants: They include thaumatin, osmotin, tobacco major and minor PR proteins, alpha-amylase/trypsin inhibitor, and P21 and PWIR2 soybean and wheat leaf proteins. The proteins are involved in systematically-acquired stress resistance and stress responses in plants, although their precise role is unknown. Thaumatin is an intensely sweet-tasting protein (on a molar basis about 100,000 times as sweet as sucrose) found in the fruit of the West African plant Thaumatococcus daniellii: it is induced by attack by viroids, which are single-stranded unencapsulated RNA molecules that do not code for protein. The thaumatin protein I consists of a single polypeptide chain of 207 residues. Like other PR proteins, thaumatin is predicted to have a mainly beta structure, with a high content of beta-turns and little helix.
Pharmaceutical manufacturing is the process of industrial-scale synthesis of pharmaceutical drugs as part of the pharmaceutical industry. The process of drug manufacturing can be broken down into a series of unit operations, such as milling, granulation, coating, tablet pressing, and others.

== History == Codeine, or 3-methylmorphine, is an alkaloid found in the opium poppy, Papaver somniferum var. album, a plant in the family Papaveraceae. Opium poppy has been cultivated and utilized throughout human history for a variety of medicinal (analgesic, anti-tussive and anti-diarrheal) and hypnotic properties linked to the diversity of its active components, which include morphine, codeine and papaverine. Codeine is found in concentrations of 1% to 3% in opium prepared by the latex method from unripe pods of Papaver somniferum. The name codeine is derived from the Ancient Greek κώδεια (kṓdeia, "poppy head"). The relative proportion of codeine to morphine, the most common opium alkaloid at 4% to 23%, tends to be somewhat higher in the poppy straw method of preparing opium alkaloids. Until the beginning of the 19th century, raw opium was used in diverse preparations known as laudanum (see Thomas de Quincey's Confessions of an English Opium-Eater, 1821) and paregoric elixirs, several which were popular in England since the beginning of the 18th century; the original preparation seems to have been elaborated in Leiden, the Netherlands around 1715 by a chemist Jakob Le Mort; in 1721 the London Pharmacopoeia mentions an Elixir Asthmaticum, replaced by the term Elixir Paregoricum ("pain soother") in 1746. The progressive isolation of opium's several active components opened the path to improved selectivity and safety of the opiates-based pharmacopeia. Morphine had already been isolated in Germany by Friedrich Sertürner in 1804.
On 26 August 1807, Bavaria introduced a similar measure. Baden followed in 1809, Prussia in 1815, Württemberg in 1818, Sweden in 1816, England in 1867 and the German Empire in 1874 through the Reichs Vaccination Act. In Lutheran Sweden, the Protestant clergy played a pioneering role in voluntary smallpox vaccination as early as 1800. The first vaccination was carried out in Liechtenstein in 1801, and from 1812 it was mandatory to vaccinate. The question of who first tried cowpox inoculation/vaccination cannot be answered with certainty. Most, but still limited, information is available for Benjamin Jesty, Peter Plett and John Fewster. In 1774 Jesty, a farmer of Yetminster in Dorset, observing that the two milkmaids living with his family were immune to smallpox, inoculated his family with cowpox to protect them from smallpox. He attracted a certain amount of local criticism and ridicule at the time then interest waned. Attention was later drawn to Jesty, and he was brought to London in 1802 by critics jealous of Jenner's prominence at a time when he was applying to Parliament for financial reward. During 1790–92 Peter Plett, a teacher from Holstein, reported limited results of cowpox inoculation to the Medical Faculty of Kiel University. However, the Faculty favoured variolation and took no action. John Fewster, a surgeon friend of Jenner's from nearby Thornbury, discussed the possibility of cowpox inoculation at meetings as early as 1765. He may have done some cowpox inoculations in 1796 at about the same time that Jenner vaccinated Phipps.
A topical source of arsenic are the green pigments once popular in wallpapers, e.g. Paris green. A variety of illness have been blamed on this compound, although toxicity has been exaggerated. Trimethylarsine, once known as Gosio's gas, is an intensely malodorous organoarsenic compound that is produced by microbial action on inorganic arsenic minerals. Arsenic(V) compounds are easily reduced to arsenic(III) and could have served as an electron acceptor on the early Earth. Lakes that contain a substantial amount of dissolved inorganic arsenic, harbor arsenic-tolerant biota.
=== 1988 Tripartite Accord === Despite taking the necessary countermeasures on the battlefield, the South African government discerned it had reached the political limits of further escalation in Angola. The casualties sustained during the Cuito Cuanavale campaign had been sufficient to cause public alarm and provoke difficult questions about the tactical situation on the border and why South African soldiers were dying there. There was little reason to believe yet another bloody campaign would be successful in expelling the Soviets and Cuba from the region; on the contrary, as in the past, it could lead to an increase in the amount of Soviet weapons and Cuban troops. The conflict had also evolved from a low-intensity struggle against lightly armed insurgents into protracted battles between armies backed by all the paraphernalia of modern conventional warfare, with the accompanying rise in human and material costs. This contributed to a sense of war weariness and increased the growing skepticism and sensitivity in civilian circles towards the SADF's Angolan operations. The failure of the Soviet-supervised Operation Saluting October, along with the consequent destruction of hundreds of millions of dollars' of FAPLA's Soviet-supplied arms, had the effect of moderating Moscow's stance on Angola. In a notable departure from its previous foreign policy stance, the Soviet Union disclosed it too was weary of the Angolan and South West African conflicts and was prepared to assist in a peace process—even one conducted on the basis of Cuban linkage.
=== 2026 capture of Nicolás Maduro and interim presidency of Delcy Rodríguez === On 3 January 2026, United States forces carried out a large-scale operation in Venezuela that resulted in the capture of President Nicolás Maduro and his wife, Cilia Flores. More than 150 aircraft were involved in the attack, and Maduro was subsequently flown to New York to face long-standing U.S. indictments on drug trafficking and weapons charges. U.S. President Donald Trump declared that the United States would "run" Venezuela until what he termed a safe transition and warned that further strikes were possible if the new authorities did not cooperate. The operation's legality was widely questioned at the United Nations and by international-law experts. Shortly after the operation, the Supreme Tribunal of Justice ordered executive vice-president Delcy Rodríguez to assume the presidency on an interim basis. The armed forces publicly endorsed her as acting head of state. Initially Delcy Rodríguez condemned the U.S. action as an "illegal kidnapping", but within days she signalled a willingness to cooperate with Washington on Venezuela's future, in a shift reported by international media. The United States has stated that it does not recognize Rodríguez as a democratically elected leader and has called for a transition culminating in "free and fair elections". Venezuelan congressman and son of Nicolás Maduro Nicolás Maduro Guerra declared on 27 January that elections in Venezuela are "not on the table" as his father was "kidnapped".

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
